DNA Sequencing Service

We offer a fast and reliable service, using ABI capillary sequencers, at competitive prices:

• DNA: PCR, Plasmid, BAC/PAC/Cosmid

• Colonies: Plasmid, BAC/PAC/Cosmid

• Quality: service up to 900 bp (500 bp - PHRED 20 guaranteed, dependent upon concentration and quality of template and primers provided)

• Turnover time: 2 working days. Submit samples till 2pm on Mondays or Wednesdays and we will email you results during following Wednesdays or Fridays respectively.

• Results are provided in any of the following formats: ab1- trace, FASTA, text, PHRED-trace or PHRED generated . Ab1 files containing sequencing traces can be opened with Chromas - please download it here: http://www.technelysium.com.au/chromas.html.

• Price: £10 for sequencing of purified DNA; £15 for sequencing of DNA from colonies

• Primers: order specific primers (add £10/primer) or enclose your own: alternatively, use standard vector primers for free

 Step 4: Assembly and shipment

• DNA (air-dried, or in 5 mM Tris-Cl pH 8-9) must be submitted in 96-well PCR plates, or 0.2 ml individual tubes or strips, labelled with name or code of template. A picture of purified plasmid or PCR product together with reference DNA (quantified DNA ladder) with legend is required for quality and quantity evaluation. Please enclose this with the samples.
• Custom primers must be submitted in coloured 0.2 ml tubes. Standard primers are available free of charge. Non-standard sequencing primers can be synthesised at cost £10. Customers provide the primer sequence or authorise us to do primer design (free of charge).
• Colonies must be submitted in 0.2 ml individual tubes or strips, or 96-well PCR plates in freezing broth (LB, containing 20% glycerol) containing antibiotic. Simply inoculate a toothpicked colony into 100 µl of broth in sterile conditions.
• Shipment: send your samples at ambient temperature in a zip-lock bag inside bubble-wrapped envelopes to Yorkshire Bioscience at ambient temperature with First Class Royal Mail or Express Courier.
