Choosing the right buffer for a particular DNA polymerase and specific application plays a key role in PCR optimisation. Poor performance of enzymes, low or no yield are consequences of employing the wrong buffer, in most cases. We offer both individual and sets of 10 × PCR buffers which have emerged during history of PCR for screening purposes. Buffers are prepared using DNase- and RNase-free water. The pH was measured at 25 ºC. All buffers were assayed in reactions with a number of commercially available DNA polymerases. Detergent-free buffers suitable for DHPLC are highlighted.
All buffers can be supplemented with MgCl2, MgSO4, or Mg-Acetate upon request.
